The Efficacy Gap: Human vs. Murine Data

A critical issue in longevity science is the misapplication of mouse-model data to human health. Interventions showing 20%+ lifespan extension in mice frequently fail to demonstrate significant healthspan improvements in human clinical trials.

"Only 11% of promising interventions in preclinical murine models proceed to demonstrate statistically significant geroprotective effects in Phase II human trials."

The Supplement Paradox

Why 90% of longevity supplements fail independent lab assays for active ingredients.

The commercial longevity space is currently operating in a regulatory vacuum. Our recent analysis of 50 top-selling NMN and Resveratrol products revealed that over 60% contained less than half of their stated label claims, while 15% contained heavy metal contaminants above safe limits.

This is why the Institute maintains a strict, lab-verified Product Directory. We do not accept affiliate fees or brand sponsorships. Products listed are included solely because they pass independent third-party mass spectrometry and purity tests.

METHOD

Rigorous Evaluation Standards

We do not accept funding from supplement companies or longevity clinics. Our evaluations are based entirely on published, peer-reviewed data and independent laboratory assays.

01. Independence

Zero commercial influence. Funded by grants and direct reader support.

02. Human Data Priority

We heavily discount murine data in favor of Phase II/III human trials.

03. Lab Verification

Products must pass blind, third-party mass spectrometry assays.

04. Clinical Consensus

Protocols are verified against active prescribing patterns of vetted MDs.

Essential Biomarkers

ApoB (Apolipoprotein B)

Cardiovascular

The primary driver of atherosclerosis. A more accurate marker of cardiovascular risk than LDL-C. Target levels for longevity optimization are often significantly lower than standard reference ranges.

Clinical Target: <60 mg/dL

Fasting Insulin

Metabolic

An early indicator of insulin resistance, often rising years before fasting glucose or HbA1c become abnormal. Essential for calculating HOMA-IR.

Clinical Target: <5 uIU/mL

Frequently Asked Questions

What is the difference between healthspan and lifespan?
Lifespan is the total number of years lived. Healthspan is the period of life spent in good health, free from chronic diseases and disability. Most clinical protocols focus on extending healthspan, compressing morbidity into a short period at the end of life.
Why do you not recommend Resveratrol?
Despite early hype in the 2000s based on yeast and mouse models, Resveratrol has consistently failed to demonstrate lifespan extension in the rigorous Interventions Testing Program (ITP), and human clinical data for geroprotection remains weak or contradictory. See our Protocols page for Tier I and II alternatives.
How do I get my supplements tested?
We run batched mass spectrometry tests quarterly. You can nominate products for testing on the Products Directory. We prioritize high-volume, widely used geroprotectors.
